Developing sex hormones such as 17-hydroxy progesterone ELISA kit, using competition and sandwich principles flexibly
17-hydroxyprogesterone Detection Kit
17-hydroxyprogesterone ( 17-OHP) is an endogenous hormone secreted by the adrenal and gonadal. It is mainly converted by progesterone or 17- hydroxypregnenolone. 17-OHP plays an important role in organ development.
17 hydroxyl progesterone content in serum is the main indicator for the diagnosis of congenital adrenal hyperplasia. The current method to detect 17 hydroxyl progesterone includes: radioimmunoassay, immunofluorescence, mass spectrometry and enzyme-linked immunosorbent assay. Compared to immunofluorescence and mass spectrometry in high demands of equipment, enzyme linked immunosorbent assay (ELISA) has advantages of harmlessness and easy operation .
By utilizing unique technology of small molecule modification and coupling, Cloud-Clone Corp. has developed 17-OHP competitive inhibition ELISA Kit (cE90454Ge). Difficulties in developing this product is mainly reflected in the preparation of immunogen and antibody. As 17 hydroxyl progesterone is a small-molecule compound (Figure 1), it has no immunogenicity. We need to modify the molecule and couple it with carrier proteins to make it immunogenic. Then monoclonal antibody with high specificity can be obtained through hybridoma technology.
Meanwhile, as a small molecule, sandwich ELISA is not applicable for the measurement of 17 hydroxyl progesterone. Therefore, we have selected the method of competitive inhibition and developed ELISA Kit for 17 hydroxyl progesterone, please see figure 2 for the standard curve of this ELISA Kit.
Sex hormones ELISA Kit
Sex hormones are divided into two categories, one of them includes 17 hydroxy progesterone, estrogen, testosterone, prostaglandin and small molecule such as ketone, alcohol and lipids. All these molecules have the same structure in different species, that is to say, they have no species specificity. Therefore, ELISA Kits for all these molecules are universal in different organisms (Ge), which can be used for quantitative detection of samples from different organisms.
Another category includes luteinizing hormone (LH) and follicle stimulating hormone (cFSH) as the representative of protein hormones. Due to high species specificity, they have unique sequences in different species. Therefore, ELISA Kits have been developed according the characteristics of this category of sex hormones.
For sex hormone series ELISA Kits developed by Cloud-Clone Corp., the minimum detection value is 12 pg/ml. They can be used for detection of lower levels of sex hormones in rat, mouse as well as unconventional samples (such as saliva, urine or tissue samples).
