BNP-an Important Biomarker of Heart Failure

 My name is B-type natriuretic peptide, also called Brain Natriuretic Peptide (abbreviated BNP). Everyone calls me that since I was found in pig brain. I am mainly derived from the ventricle. In order to make everyone to know more about me, first of all, let me talk about my experience. Human cardiac cells secrete B-type natriuretic peptide (His27-His134), when ventricular volume overloads or pressure load increases, some endonucleases cleave B-type natriuretic peptide to form N-terminal B-type natriuretic peptide (NT-proBNP, His27-Arg102) and me (BNP, Ser103-His134). I have biological activities and play multiple roles in natriuretic, diuretic, antihypertensive and smooth muscle relaxation processes through binding to certain target cell surface receptors, thus I has become a vital biological cardiovascular indicator. Furthermore, nowadays heart failure (especially early heart failure) is difficult to diagnose, and some patients do not have typical symptoms of heart failure (including polypnea, ankle swelling, fatigue, etc.). Sometimes it is difficult for physicians to diagnose whether these symptoms are result from cardiac dysfunction or not, especially for elderly patients and obese patients. On the other hand, routine detection methods for heart failure, including echocardiography, radionuclide imaging and cardiac MRI, are all prone to false positives, and the repeatability of these methods is not very well, too. As one of important heart failure biomarkers, many researchers expect to understand me better. According to the BNP amino acid sequence on the website of Uniprot, researchers from Cloud-Clone Corp. developed human BNP peptide, and the sequence is shown in Fig.1. Then, they obtained synthetic peptide BNP (Cat # CPA541Hu01).

Since I am too short (only 32aa) that I don’t possess the immunogenicity, people cannot produce antibodies against me by directly animal immunization. The R&D team of Cloud-Clone Corp. solved this challenge perfectly. They developed OVA-BNP (Cat # CPA541Hu21) by using a new molecular transformation and coupling technology. Then after immunizing animal, they obtained rabbit polyclonal antibody against human BNP (Cat # PAA541Hu02) and mouse anti-human BNP monoclonal antibody (Cat # MAA541Hu22).

Cloud-Clone Corp. then labeled purified polyclonal antibody (Cat # PAA541Hu02) with Biotin or FITC, respectively, and obtained Biotin-Linked Antibody to Brain Natriuretic Peptide (Cat # PAA541Hu71) and FITC-Linked Antibody to Brain Natriuretic Peptide (Cat # PAA541Hu81). And the IHC image in heart muscle tissue (Fig.2) shows that there is a strong signal in human cardiac tissue, confirming that this antibody has high specificity.

Since traditional sandwich ELISA strategy is not suitable for detecting of small molecules. Cloud-Clone Corp. developed a competitive ELISA that have much higher specificity and sensitivity (Cat # CEA541Hu) to quantify my level in biological samples.
