 An autoantibody is an antibody (a type of protein) produced by the immune system that is directed against one or more of the individual's own tissues, organs,cells and cellular components (including proteins and nucleic acids). Normally, immune cells can discriminate self from non-self. A normal body does not respond to its own antigens. This self-defense mechanism of immune system is called Immune Tolerance. The body maintain normal growth, development and survival by immune tolerance systerm. Once this system is destroyed, the immune system recognize one or more of the body's normal constituents as "non-self", and leading to production of pathological autoantibodies. Serum from healthy individuals may contain low level autoantibodies. Low level autoimmunity may not lead to diseases,while a high level of autommunity is unhealthy. The autoantibodies may attack the body's own healthy cells, tissues, and/or organs, causing inflammation, damage and autoimmune diseases, such as Systemic lupus erythematosus(SLE), rheumatoid arthritis (RA) and so on. Autoantibodies are used as an important tool to diagnose many autoimmune diseases. Along with the development of Early diagnosis and Standardized treatment, the role of autoantibodies in the diagnosis, monitoring and prognosis increasingly draw attention from biomedical field.

Currently, the pathogenesis of autoimmune diseases is still entirely clear and the exact meaning of some autoantibodies in disease has not yet been rigorously proven, but the correlation have been recognized. And the presence of autoantibodies is one of the basic features of the autoimmune disease. Currently, the autoantibodies are the most widely used clinical diagnosis indicators for autoimmune disease since the autoantibodies exist in serum of patients with autoimmune disease in most cases. Moreover, autoantibodies monitoring will be benefit to auxiliary diagnosis, clinical judgment and therapeutic effect observation of autoimmune disease.

Instead of common used techniques, such as Indirect Immunofluorescence Assay(IIF), Immunodiffusion Assay(ID), Western-blot(WB) and so on, ELISA and CLIA are the best choices to detect autoantibodies recently, since the two methods have various advantages, including high sensitivity, specificity indexes, high speed, low cost, easy to operate and so on.
