The preparation of rat adjuvant arthritis model

Rheumatoid arthritis is a chronic autoimmune disease that is associated with complex inflammatory mediators, which will cause joint injury,synovial inflammation, cartilage and bone damage.The typical characteristic of this chronic disease is the chronic joint injury, seriously affecting people's health and the development of animal husbandry.

  The incidence of RA is generally believed to be caused by multiple factors,which include genetic factors,bacteria and virus mediated infection factors,autoimmune tolerance damage, endocrine disorders and other factors of cold,damp, fatigue, trauma, smoking and mental stimulation.The pathological symptoms were mainly manifested in the aspects of immune function disorder, proliferation and thickening of articular synovial tissue, cartilage and bone destruction and so on.Many studies have shown that lymphocytes, neutrophils, giant salivary cells, synovial cells and other inflammatory cells mediated the abnormal cellular and humoral immunity, leading to the joint local and systemic immune dysfunction which are important cause for the RA.In addition, some cytokines such as tumor necrosis factor alpha, interleukin 1 and interleukin 8 play an important role in the occurrence and development processes.At present, although a lot of studies have been done on RA, its pathogenesis is still not clear.

  Due to the limitation of human ethics, law and so on, people can not directly use human as the research object in some medical fields. So, some other animal models must be used for the study. The studies have shown that the rat adjuvant arthritis model have many similar characteristics with the human RA in the clinical manifestations, path- ological, immunological and pathological mechanism, etc. So it is an ideal animal model to study the pathological mechanism of RA and evaluate the treatment of RA drugs.
