Jam4 and MAGI1 interacted with each other and formed clusters in transfected COS-7 cells, and Jam4 mediated calcium-independent homophilic adhesion and was accumulated at cell-cell contacts when expressed in mouse fibroblasts. MAGI1, ZO1, and occludin were recruited to Jam4-based cell contacts. Jam4 also reduced the permeability of Chinese hamster ovary (CHO) cell monolayers. MAGI1 strengthened Jam4-mediated cell adhesion in mouse fibroblasts and sealing effects in CHO cells.
The deduced 370-amino acid protein has a calculated molecular mass of 40 kD. It has an N-terminal signal peptide, followed by 2 V-type Ig loops, a transmembrane domain, and a C-terminal PDZ-binding motif.
