This gene is orthologous to mouse HDAC7 gene whose protein promotes repression mediated via transcriptional corepressor SMRT. Multiple alternatively spliced transcript variants encoding several isoforms have been found for this gene. Dot blot analysis of more than 40 human tissues showed that highest expression of HDAC7 was in adult and fetal thymus. In situ hybridization analysis revealed most abundant expression of HDAC7 in the cortical region of thymus. Flow cytometric analysis demonstrated highest HDAC7 expression during the double-positive stage of thymocyte maturation. either the amino or the carboxyl terminus of HDAC7 is sufficient for transcriptional repression and that the repression of HDAC7 is insensitive to trichostatin A, indicating that HDAC7 represses Mitf at least in part by deacetylation-independent mechanism.
