The deduced 1,732-amino acid PKD1L3 protein contains an N-terminal C-type lectin domain, followed by a central GPCR proteolytic site and PLAT domain, a pair of PKD domains, and a C-terminal putative channel pore region. It also has 11 transmembrane domains.
PKD1L3 does not possess the receptor egg jelly domain found in PKD1L2. Confocal microscopy and coimmunoprecipitation analysis revealed coexpression of Pkd1l3 and Pkd2l1, but not Trpm5, in the apical ends of taste cells in circumvallate and foliate papillae. Coexpression of Pkd1l3 and Pkd2l1 was necessary for inducing changes in intracellular calcium concentration in response to acid solutions, suggesting that PKD1L3/PKD2L1 heteromers function as sour taste receptors.
