2-Aminothiazole (AT)
2-Aminothiazole is a heterocyclic amine with odor similar to pyridine, soluble in water, alcohol and ether. It is a beginning point for synthesis of many compounds including sulfur drugs, biocides, fungicides, dyes and chemical reaction accelerators. 2-Aminothiazole can be used as a thyroid inhibitor in the treatment of hyperthyroidism and it has antibacterial activity. Also used as the acid tartrate. A 2-aminothiazole derivative 1 was developed as a potential inhibitor of the oncology target AKT, a serine/threonine kinase. When incubated in rat and human liver microsomes in the presence of NADPH, 1 underwent significant metabolic activation on its 2-aminothiazole ring, leading to substantial covalent protein binding. Upon addition of glutathione, covalent binding was reduced significantly, and multiple glutathione adducts were detected.
