Zinc Finger Protein 148 (ZNF148)
ZNF148 inhibited SP1 activation of the ornithine decarboxylase (ODC) gene. ZNF148 and SP1 bound to a GC box in the proximal promoter of the ODC gene in a mutually exclusive manner. ZNF148 also repressed SP1-mediated activation of the vimentin promoter. The N-terminal zinc fingers of ZNF148 interacted with SP1 and repressed gene expression, and cotransfection of TAFII130 (TAF4) with ZNF148 and SP1 in insect cells rescued expression.ZNF148 gene contains 9 exons and spans about 130 kb. Exon 4 contains the translation initiation site. Mouse Znf148 showed the same genomic structure with identical intron-exon organization, and the promoter region displayed typical features of a housekeeping gene, with a high G/C content and the absence of canonical TATA and CAAT boxes.
