USP33 is a widely expressed, deubiquitinating enzyme first identified as a von Hippel-Lindau tumor suppressor (VHL) protein-interacting deubiquitinating enzyme (hVDU1) and is a member of the peptidase C19 family. USP33 is involved in a variety of processes including beta-2 adrenergic receptor/ADRB2 recycling and cell migration. USP33 is a required component of the Slit signaling pathway responsiveness in breast cancer cells. Slit redistributes Robo1 to the plasma membrane from intracellular compartments in a manner dependent on the presence UP33, which produces an inhibitory effect on breast cancer cell directional migration. USP33 has been shown to bind to and deubiquitinate D2 as a crucial response to cold exposure, helping to decrease D2 degradation, and acclimatize brown adipocytes.
