- Modeling Method1. Fasting for one night before the operation, free drinking water.
2. Weight the rats and anesthetized by intraperitoneal injection of chloral hydrate (15%) 350mg/kg, shaving and use iodine and alcohol to wipe operation area.
3. Cut into the wall in the abdominal midline left about 3-4mm, seperate the left kidney and ureter for modeling group rats
4. In the near renal pole, ligation and separation of the ureter and double ligation, in the two intermediate ligation cut ureter, layer by layer suture muscle and abdominal wall, alcohol to clean the suture. After the rats awake, put it back into the cage and put it back into the feeding room, observe the state and the death of the rat and make a record.
5. Operated rats are placed in the heating pad to its wake up. The sham operation group did not ligation of the ureter, and the other steps are the same.
6.Injection of penicillin 200 thousand units/day to prevent infection for three days after operation.
Note:
1. Don not damage the kidneys when separation of the ureter.
2. Should clean up the wound to prevent the infection.

Model Evaluation
1.Urine protein and Urine NAG activity determination
Urinary protein and urinary NAG activity of the model group is significantly higher than that of the normal group and the sham operation group at 2 and 4 weeks after operation.
2. Serum creatinine (Scr), Urea nitrogen (BUN) content is determined after 4 weeks of operation, the model group serum Scr and BUN levels is increased.
Pathological Results
The left kidney is fixed with 4% poly formaldehyde, dehydrated, transparent, paraffin embedded and sliced. By HE staining and Masson staining, the pathological changes and inflammatory cell infiltration of glomerular and renal tubular interstitial are observed by light microscope.

Statistical Analysis
SPSS software is used for statistical analysis, measurement data to mean ± standard deviation (x ±s), using t test and single factor analysis of variance for group comparison, P<0.05 indicates there was a significant difference, P<0.01 indicates there are very significant differences.
